Part 1: Diagnosis and risk stratification of brain lesions is challenging when tissue biopsy is contraindicated due to anatomic or clinical factors.
Cerebrospinal fluid (CSF)–based liquid biopsy using cell-free DNA (cfDNA) and RNA (cfRNA) offers a minimally invasive approach to detect
neoplastic disease and inform management. We evaluated the clinical utility of a commercially available cfDNA/cfRNA next-generation sequencing
assay applied to CSF specimens from patients with central nervous system (CNS) lesions of uncertain etiology.
Part 2: Enumeration and detection of HER biomarker status on circulating tumor cells (CTCs) in cerebrospinal fluid (CSF) has clinically actionable insights
that could expand therapeutic options for patients with leptomeningeal metastasis (LM). Done in combination for select patients, information derived from
cell free DNA/RNA (cfDNA/RNA) analysis could provide early indicators of developing tumor resistance to systemic therapies and inform therapeutic pivot
for improved patient outcomes.
